Sop for operation and cleaning of Dynamic pass box.

I. Purpose & Scope:

  • The purpose of this SOP is to lay down a procedure for operation and cleaning of Dynamic pass box used in Microbiology laboratory
  • This Standard operating procedure shall be applicable for operation and cleaning of Dynamic Pass box in the Microbiology section of Quality Control Department

II. Responsibilities:

  • All Quality Control and Quality Assurance personnel’s shall be responsible to follow and implement this SOP.

III: Introduction and Operation :

  • Pass box helps to prevent the air flow from one area to another, while transferring the material.
  • One door of the pass box opens at a time and both doors shall not be open simultaneously.
  • Dynamic pass box works like an air-lock. Clean air enters in the pass box through blower and it prevents the entry of external air into the pass box.
  • Ultra-violet (UV) light is provided in the pass box to reduce the microbial contamination that may enter in the pass box during the transfer of material.

Operation of Dynamic Pass Box  :

  • Switch ‘ON’ the ‘Mains’ power supply of Dynamic Pass Box. Display will show ‘Crescendo India’, Then UV lamp will start automatically, now display will show the status as ‘UV light is ON’ and total UV burning hours will be indicated as ‘Hours’.
  • Open the door by pressing the ‘OPEN’ button present near the display. Once the button is pressed the display will show ‘Open the door within 5 seconds’, and seconds will start to blink. Then open the door within 5 seconds by pulling it out.
  • Once the door is opened, blower will start and UV lamp will be off automatically. Ensure that the differential pressure should be in between 7.5 to 13 mm of water.
  • Before opening the door, ensure that opposite door is closed.
  • After opening the door, ensure that UV light is off and blower is on.
  • Keep the material to be transferred in the Pass box.
  • Close the door of pass box and ensure that UV light is switched on.
  • Allow to expose the material with UV light for appropriate time.
  • Open the opposite side door of the pass box and unload the material.

Cleaning :

  • Clean the dynamic pass box using 70% IPA, with the help of lint free cloth.
  • Open the door of Pass box.
  • Ensure that the blower is on and UV light is off.
  • Clean entire surface of pass box with the filtered 70% IPA solution by using lint free cloth. Clean inside and outside of the pass box, before and after usage.
  • Wipe the UV tube lamp, with dry lint free cloth. Do not spray 70% IPA directly on UV tube lamp.

Precautions :

  • Record the reading of UV hours once in a day, in ‘UV lamp burning record of Dynamic pass box’ in Annexure-I.
  • UV lamp should be replaced after burning for 5000 hours.
  • During transfer of the microbial cultures, positive controls and samples to be tested through the pass box, ensure that the UV light is switched off.
