Sop for Operation and Cleaning of Lacquer Porosity Apparatus

I. Purpose & Scope:

  • The purpose of this SOP is to lay down the procedure for Operation and Cleaning of Lacquer Porosity Apparatus
  • This Standard operating procedure shall be applicable for Operation and Cleaning of Lacquer Porosity Apparatus in the Microbiology section of Quality Control Department

II. Responsibilities:

• All Quality control personnel shall be responsible to follow and implement this SOP.

III : Introduction and Procedural Part :

General cleaning of Lacquer Porosity Apparatus :

  • Clean the instrument with a clean dry cloth everyday. Occasionally wet cloth dipped in dilute soap solution may be used.
  • Precaution has to be taken to clean the instrument immediately with dry cloth to remove the moisture.

Precautions: 

  • While testing the lacquer porosity of tube, cathode should not touch to the surface of the tube.
  • Clean the electrode after every use with polish paper.
  • Replace with correct rating of fuse whenever require.
  • Follow the safety precautions when handling chemicals like Copper Sulphate.

Basic operation of Lacquer Porosity Apparatus :

  • Switch “ON” the main power supply, & then the Switch “ON’ the “ Main” switch, of machine , the power indicator will glow indicating the apparatus is getting power supply.
  • Loosen the knurling screw on the copper electrode and its control bark elite disc and lift the electrode to its maximum position and tighten the electrode with knurled screw.
  • Fill the collapsible tube up to 1 inch form the top with 1% copper sulphate solution and hold the tube in the Stainless steel jaw on the Apparatus.
  • Loosen the knurled screw and lower down the copper electrode so as to dipped in the copper sulphate solution. Control Bakelite disc with the knob to hold the tube in proper position. Press conical disc on open end of the tube slightly and then tighten the knurled screw on the conical disc.

Check: To test the proper working of the machine, press ‘CHECK’ push button for 4 to5 second, display will shows reading between 450 to 550 mAmp.

TEST : For performing test press TEST push button about 5 second; displays will show test reading. Record the results as observed & check the requirements as

  • After taking reading, lift the lever and bring it to its original position. Cut the collapsible tube by scissor in a lengthwise direction and open the inside lacquer coated surface to the tube for visual observation, Copper sulphate will be seen deposited, wherever there is lacquer puncture on the inside coating of lacquer in the collapsible tube.
  • Preparation of Copper Sulphate solution as follows,

Take & weigh the following materials in 100 ml volumetric flask & dilute to 100 ml with distilled water.

 Calibration :

  • Calibration Frequency: Yearly
  • Calibrate the apparatus from the outside party once in a year.
  • Handle the out of calibration results as per specified Sop
  • Note: If apparatus does not comply with any of the calibration parameters then follow the relevant procedure.
